چکیده

Hepatomas were induced in monkeys by the administration of jV-nitrosodiethylamine. An a-fetoprotein, not normally found in adult monkeys, was present in the serum of the majority of the animals with the liver tumor. Frequently, the test for serum a-fetoprotein was positive prior to any histolÃ3gica!evidence of a cancer. With tRNA from Escherichia coli K12W6 or from normal adult monkey liver or hepatoma as methyl acceptor, an increasing degree of methylation was found with the tRNA methylases in tissue extracts from normal adult monkey liver, premalignant liver, liver from newborn monkeys, and hepatoma, respectively. These observations suggest that a change in specificity of tRNA methylation occurs during chemical carcinogenesis with Af-nitrosodiethylamine. The increase in tRNA methylation by extracts of premalignant liver also suggests that changes in tRNA methylase activity occur prior to, or as an early manifestation of, malignant conversion. Further study showed the presence of an inhibitor of tRNA methylases in extracts from normal adult monkey liver but not in extracts of normal newborn liver and hepatoma. Thus, multiple factors may be involved which account for the differences observed in the tRNA methylases.

Increased transfer RNA methylase activity in a liver tumor from chicks with Marek's disease.

virus was increased up to 200 percent compared to uninfected animals. This elevation in enzyme activity occurred after the appearance of discrete liver tumors and was highest in isolated tumor nodules which had been separated from adjacent tissue. Other organs (spleen, kidney, and lung) with no visible lesions showed no changes in methylase activity. Transfer RNA isolated from tumorous liver of...

The methylation of transfer RNA in vitro by extracts of normal and malignant tissue.

The transfer RNA (tRNA) methylase activities of normal and regenerating rat liver, liver of newborn rats, and a minimum deviation hepatoma have been studied in vitro with Escherichia coli tRNA as substrate. The levels of activity are higher in extracts of malignant, newborn, and regenerating tissue than in extracts of normal tissue, when comparison is made at the optimum concentration of ammoni...

Transfer RNA methylase and transfer RNA methylase-inhibitor activity in normal and malignant human ovarian tissue.

Eight different human ovarian carcinomas were shown to have 2 to 25 times higher transfer RNA methylase-specific activity and transfer RNA methylating capacity than do normal ovarian tissues. The more rapidly metastasizing, poorly differentiated carcinomas had higher transfer RNA methylase activity than did slower metastasizing, well-dif ferentiated, and intermediately differentiated carcinomas...

Ethionine-induced changes in rat liver transfer RNA methylation.

We have confirmed the finding by Rajalakshmi that transfer RNA (tRNA) from livers of ethionine-treated rats can act as a substrate for homologous tRNA-methylating enzymes in vitro. This methyl-deficient tRNA from liver can be methylated in vitro by enzymes from normal or ethionine-treated rats. The in vitro inhibition of tRNA methylation that follows ethionine treatment can be at least partiall...

Increase of soluble RNA methylase activities by chemical carcinogens.

High levels of the hepatocarcinogen ethionine (1% of the DL isomer mixture) in diets were shown to increase soluble RNA methylase activity of rat and hamster liver cells. Enzyme increases were detected after only 4 days of ethionine feeding. Dimethylaminoazobenzene, another hep atocarcinogen, also increased soluble RNA methylase activ ity. Noncarcinogenic analogs of ethionine, norleucine and L-...
